Synthesis of 2'-O-methoxyethylguanosine using a novel silicon-based protecting group.
A short and efficient synthesis of 2'-O-methoxyethylguanosine (8) is described. Central to this strategy is the development of a novel silicon-based protecting group (MDPSCl(2), 2) used to protect the 3',5'-hydroxyl groups of the ribose. Silylation of guanosine with 2 proceeded with excellent regioselectivity and in 79% yield. Alkylation of the 2'-hydroxyl group of 6 proceeded with methoxyethyl...
متن کاملChemical synthesis of a very long oligoribonucleotide with 2-cyanoethoxymethyl (CEM) as the 2′-O-protecting group: structural identification and biological activity of a synthetic 110mer precursor-microRNA candidate
A long RNA oligomer, a 110mer with the sequence of a precursor-microRNA candidate, has been chemically synthesized in a single synthesizer run by means of standard automated phosphoramidite chemistry. The synthetic method involved the use of 2-cyanoethoxymethyl (CEM), a 2'-hydroxyl protecting group recently developed in our laboratory. We improved the methodology, introducing better coupling an...
متن کاملA Novel Method for the Synthesis of 2-Ketomethylquinolines
Several 2-ketomethylquinolines were synthesized by heating, 2-methyl quinoline with acyl chloride in the presence of AlCl3 Contrary to reported procedures, this method was accomplished at a short time obtaining good results.
متن کامل2-Benzamidoethyl group--a novel type of phosphate protecting group for oligonucleotide synthesis.
A number of 5'-O-(4,4'-dimethoxytrityl)thymidine N,N-diisopropylamino phosphoramidites protected at P(III) with derivatives of 2-benzamidoethanol were synthesized and incorporated into synthetic oligonucleotides. Depending on substitution patterns at the alkyl chain, amido group, and phenyl ring, the time required for removal of these protecting groups using concentrated ammonium hydroxide vari...
متن کامل2 Synthetic methods Part ( iii ) Protecting groups
Another excellent and comprehensive annual ‘update’ review of the 2000 literature relating to protecting group (PG) strategies in organic synthesis has appeared this year. The employment of PGs to direct ‘docking’ at oxidative enzyme active sites thereby controlling subsequent hydroxylation of unactivated carbon atoms has also been overviewed. Moreover, a summary of the use of microwave (MW) he...
